摘  要:目的探讨叶黄素对体外培养的人食管癌9706细胞(esophageal carcinoma 9706,Ec-9706)增殖和细胞凋亡的影响。方法将Ec-9706细胞用含10%胎牛血清的RPMIl640培养基于37℃、5%CO_2/95%空气饱和湿度培养箱中培养。采用随机数字表法,将其随机分为4组:对照组(C组)和不同浓度叶黄素组(L_(1-3)组)。L_(1-3)组分别加入40、80和160μmol/L叶黄素;C组加入等体积RPMI1640培养液。采用噻唑兰法检测不同浓度叶黄素对人食管癌Ec-9706细胞增殖的影响;采用流式细胞术检测Ec-9706细胞凋亡情况;采用免疫细胞化学方法检测Fas、增殖细胞核抗原(proliferating cell nuclear antigen,PCNA)、cyclin D1和Caspase-3蛋白的表达。结果叶黄素可抑制Ec-9706细胞的增殖,诱导Ec-9706细胞的凋亡,且呈剂量依赖性。叶黄素可下调Ec-9706细胞PCNA和cyclin D1蛋白表达,上调Fas和Caspase-3蛋白表达,且呈剂量依赖性。结论叶黄素可抑制体外培养的人食管癌Ec-9706细胞的增殖,诱导其凋亡,且呈浓度依赖性,其机制与下调PCNA和cyclin D1蛋白表达,上调Fas和Caspase-3蛋白表达有关。Objective To observe the effects of lutein on the human esophageal carcinoma 9706 (Ec-9706) cell line in vitro. Methods proliferation and apoptosis of The inhibitory effect of lutein on the proliferation of human Ec-9706 cells at different concentrations was determined by methyl thiazolyltetrazolium(MTT) assay. The changes of cell apoptosis rate of tumor cells were determined by flow cytometry (FCM). The expression of Fas, proliferating cell nuclear antigen ( PCNA), cyclin D1 and Caspase-3 proteins were examined by immunocytochemical technique. Results Lutein significantly inhibited the proliferation and induced the apoptosis of Ec-9706 cells in a dose-dependent manner. Lutein dramatically inhibited protein expression of PCNA and cyclin D1 ,while up-regulated the expression of Fas and Caspase-3 protein. Conclusion Lutein can inhibit the proliferation of Ec-9706 cells and induce the apoptosis of Ec-9706 cells in vitro by down-regulating the expression of PCNA and cyclin D1 and upregulating Fas and Caspase-3 expression.
