摘 要:目的:观察荷乳腺癌小鼠电针后其视交叉上核(SCN)mPer1、mPer2、mCry1、mCry2表达水平的变化,探讨电针对荷瘤动物自发性活动节律影响的生物钟负反馈基因调控机制。方法:将18只C57雄性小鼠随机分为空白组、模型组、电针组3组,每组6只。LD驯化10 d后关闭光源使动物处于自由运行状态,DD驯化10 d后开始造模,造模成功继续监测节律7 d后进行电针干预。电针组在CT4针刺百会和长强穴,使用韩氏穴位神经刺激仪,频率2/15 Hz,电流0.5 mA,每次15 min,每日1次,连续电针3次。空白组和模型组在相应时相点CT4用同样的方法抓取和固定。第三次电针后2 h立即断颈处死动物,采用实时荧光定量PCR法测定各组小鼠视交叉上核(SCN)内Per1,Per2,Cry1和Cry2 mRNA表达量。结果:模型组视交叉上核Per1、Per2、Cry1和Cry2 mRNA表达水平均有降低趋势,电针组Per1、Per2、Cry1和Cry2 mRNA表达水平均有升高趋势。结论:电针对荷瘤小鼠自发性活动节律的调整作用机制可能与调控SCN负反馈基因表达有关。Objective:Observe the change of mPer1,mPer2,mCry1 and mCry2 in SCN of mice bearing breast cancer after electro-acunpuncture for further discuss on the negative feedback clock genes regulating mechanism of electro-acupuncture on the spontaneous activity rhythm of animals bearing tumors.Methods:18 male C57 mice were randomly divided into 3 groups with 6 in each group.Each group received LD domestication for 10 days.After that,light source was blocked so that the mice were in free movement and they received DD domestication for 10 days before model establishment started.Upon successful model establishment,the spontaneous activity rhythms of mice were monitored for 7 days.After that,electro-acupuncture was administered on the electro-acupuncture group.Electro-acupuncture was administered at Changqiang(GV 1) and Baihui(GV 20) on CT4 with HANS Acupoint and Nerves StimuLator(HANS-200)using 2/15Hz and 0.5 mA for 15 min each time and one time a day.Animals were immediately sacrificed 2 hours after the third time.The sample were extracted and detected by real-time PCR.Results:The Per1mRNA,Per2mRNA,Cry1mRNA and Cry2mRNA levels in SCN of model group were lower than blank group but there were no significant differences(P0.05) respectively.The Per1mRNA,Per2mRNA,Cry1mRNA and Cry2mRNA levels in SCN of electro-acupuncture group were higher than model group but there were no significant differences(P0.05).Conclusions:The regulating mechanism of electro-acupuncture on the spontaneous activity rhythm of tumor-bearing mice was probably in connection with the regulation of negative feedback clock genes expression in SCN which should be proved by further experiments.
