摘  要:为提高漆酶产量,进一步拓展漆酶开发利用空间,本文研究碳源、氮源、诱导剂、铜离子4种成分对鸡腿菇液态培养漆酶产量的影响。以ABTS为底物,用紫外分光光度法测定漆酶活性,连续测定20d左右。测定结果表明:供试碳源中,蔗糖促进鸡腿菇产漆酶效果最明显,在产酶液态基础培养基中加入2%蔗糖时,鸡腿菇漆酶产量最高达1827.5U/L;氮源中酵母粉效果最明显,在加有2%蔗糖的产酶液态培养基中加入0.2%酵母粉时,漆酶最高产量为679.1667U/L;愈创木酚在供试诱导剂中促进效果最明显,在加有2%蔗糖、0.2%酵母粉的产酶基础培养基中加入0.1mmol/L愈创木酚诱导剂时,鸡腿菇产漆酶最高达1479.792U/L;在加有2%蔗糖,0.2%酵母粉的产酶基础培养基中加入3.0mmol/L硫酸铜时,漆酶产量达到最大,为1154.167U/L。Coprinus comatus was inoculated in different liquid medium with different medium component including carbon sources, nitrogen sources, inducers or/and different concentration of Cu^2+ so as to investigate the effect of the above four medium components on laccase production by Coprinus comatus. The laccase activity was detected in the UV- spectrophotometric method used ABTS as the substrate and lasted for 20 days or so. The results showed that sucrose, yeast extract, Guaiacol and 3.0 mmol/L Cu^2+ had distinct effect on laccase production by Coprinus comatus among their counterparts. In the medium with 2% sucrose the laccase activity peak reached 1 827.5 U/L. While added 0.2% yeast extract in the medium with 2% sucrose, the laccase activity peak was 679.1667 U/L. The highest laccase activity with 1 479.792 U/L was detected in the medium of 2% sucrose, 0.2% yeast extract and 0.1 mmol/L Guaiacol. When 0.2, 0.4, 0.6, 0.8, 1.0, 3.0, 5.0 mmol/L CuSO4 was added to the medium with 2% sucrose, 0.2% yeast extract, it was found that the laccace activity reached its peak of 1 154.167 U/L in 3.0 mmol/L Cu^2+ medium. Based on the results, it is expected to contribute to further study of optimizing the production of laccase by Coprinus comatus.
