摘  要:目的:研究Zn2+沉淀法纯化柿果多酚的最佳条件及其抑菌活性。方法:采用单因素及正交试验,得到最佳沉淀及转溶条件,并检测纯化后柿果多酚对8种常见致病菌的最小抑菌浓度。结果表明:柿果多酚沉淀最佳条件为m(柿果多酚)∶m(ZnCl2)=1∶5,pH6.8,时间25min,温度30℃;其转溶最佳条件为盐酸浓度2.5mol/L,料酸比1∶5g/mL,时间20min,温度30℃。在此条件下多酚得率12%,纯度92.6%。柿果多酚对大肠杆菌、金黄色葡萄球菌、鼠伤寒氏沙门氏菌、奇异变形杆菌、志贺氏痢疾杆菌、乙型溶血链球菌、肺炎链球菌及粪肠球菌最小抑菌质量浓度分别为0.1、0.05、0.1、0.2、0.8、1.6、1.6、0.05mg/mL。结论:Zn2+沉淀法可有效纯化柿果多酚,纯化后的柿果多酚有较强抑菌活性。Objective: the optimum condition of purification of persimmon polyphenol by Zn^2+ sedimentation method and its antimicrobial activity were studied. Method: single-factor and orthogonal experiments were conducted to determined the optimum conditions of sedimentation and dissolution. The minimal inhibitory concentration of purified persimmon polyphenol on 8 common pathogenic bacteria was examined as well. Result: the optimum parameters of sedimenta- tion were rn (persimmon polyphenol):m (ZnC12)=I:5, pH 6.8, 25 min and 30 ℃; the optimum parameters of dissolution were: 2.5 moL/L HCl, m(sediment):v(HC1)=l:5(g:mL), 20min and 30℃, under this condition, the yield and purity of persimmon polyphenol could reach 12% and 92.6%, respectively. Persimmon polyphenol showed inhibitory effects on Escherichia coli, Staphylococcus aureus, Salmonella typhimurium, proteus mirabilis, Shigella dysenteriae, hemolytic streptococcus, streptococcus pneumonia and Enterococcus faecalis, with the minimal inhibitory concentration 0.1, 0.05, 0.1, 0.2, 0.8, 1.6, 1.6 and 0.05 mg/mL respectively. Conclusion: Zn^2+ sedimentation method could be an effective way to obtain purified persimmon polyphenol which possess a good antimicrobial activity.
