摘 要:以酵母细胞为材料,研究SO2衍生物(Na2SO3-NaHSO3混合液,3∶1,mmol.L-1∶mmol.L-1)致细胞死亡的信号途径.结果表明,在浓度25~100mmol.L-1范围内,SO2衍生物处理可引起酵母细胞死亡;一定浓度的抗氧化剂抗坏血酸(AsA)和Ca2+螯合剂乙二醇双四乙酸(EGTA)与SO2衍生物共同作用时,酵母细胞死亡率显著降低.研究结果表明,一定浓度的SO2可诱导酵母细胞死亡,胁迫可能通过诱导胞内活性氧和Ca2+升高,引发细胞死亡.The effects of SO2 derivatives on cells death were investigated in model eukaryote yeast.The yeast cells were treated with various chemicals,stained with methylene blue dye,and then examined with a microscope.The randomly selected views on the tested yeast cells were monitored to determine the number of dead cells and the total number of scored cells.SO2 derivatives,a mixture of sodium sulfite and sodium bisulfite(3∶1,mmol·L-1∶mol·L-1),significantly induced cell death at concentrations from 25 to 100 mmol·L-1.However,the percentage of cell death decreased when yeast cells exposed to SO2 derivatives combined with either ascorbic acid(AsA,1 mmol·L-1) or EGTA(Ca2+ chelator,0.5 mmol·L-1).The results of the present study indicated that SO2 derivatives caused cell death via reactive oxygen species(ROS) production,as well as intracellular Ca2+ level increase in yeast cells,both of them could mediate cell death.
