摘  要:用正交实验方法研究水解酶联合自溶作用获得南极磷虾水溶性多肽的优化条件,通过SDS-PAGE、凯氏定氮、氨基酸分析了南极磷虾酶解液的多肽含量、多肽分子量以及氨基酸组成。结果显示,酶解的最佳条件为使用胰蛋白酶联合自溶作用在45℃的条件下水解12h;酶解液中分子量在11.1~49.3ku的多肽最高可占水溶性南极磷虾多肽的68.01%;南极磷虾多肽的氨基酸比例均衡,不同处理条件对南极磷虾多肽的氨基酸模式影响较小,EAA/TAA和EAA/NEAA分别在40以上和64以上,符合FAO/WHO推荐的理想蛋白质模式。Optimum conditions to obtain water-soluble peptides from Antarctic Krill by protease united autolysis by orthogonal test were studied.The content,molecular mass and amino acid composition of Antarctic krill Peptid were determined by Kjeldahl method,sodium dodecyl-sulfate polyacrylamide gel eletrophoresis(SDS-PAGE) and automated amino acids analysis. Results showed that trypsin united autolysis, 12h in hydrolysis and temperature at 45℃ were optimum condition,under which hydrolyzed peptide with the molecular weight from11.1ku to 49.3ku that were up to 68.01% of total peptide and the amino acid ratio of Antarctic krill Peptide was balanced. Different treatment on Antarctic krill had a smaller impact on the result of amino acid analysis. EAA/TAA and EAA/NEAA respectly was in 40 and 64 above,in line with FAO/WHO recommended ideal protein pattern.
