摘　　要：对微生物学实验"产生蛋白酶菌株的筛选、鉴定、培养和选育"进行了教学改革,改变了以往的内容安排以及考核方式,并将该实验综合为一个统一的实验模块.模块是由8个紧密相连的实验所组成的一个综合性实验模块,其每个实验的具体内容分别为:玻璃器皿的清洗、包扎,培养基的配制及灭菌,产蛋白酶菌株的筛选、纯化及保藏,产蛋白酶菌的革兰氏染色及镜检,产蛋白酶菌的PCR鉴定,16S rDNA序列的比对及其分析,产蛋白酶细菌的生理生化鉴定,产蛋白酶菌的培养优化实验以及其诱变选育.Contents and examination forms of teaching comprehensive experiment of microbiology, which focuses on isolation, identification, cultivation and breeding of protease-producing strains, are introduced in this paper. The comprehensive experiment was composed of eight closely linked parts, which were as follows： clean and pack of glassware, and preparation and sterilization of media; screening, purification and preservation of protease-producing strains; gram stain and examination of microscope; PCR identification of strains; alignment and analysis of 16S rDNA sequence of the strains; identification of physiological and biochemical characters of the strains; optimization of the culture medium and mutation breeding of the strains.
