双自杀基因重组腺病毒对SCG7901细胞的体外杀伤作用

摘  要:目的探讨KDR启动子驱动的CD/TK双自杀基因重组腺病毒(Ad-KDRp-CDglyTK)对胃癌SCG7901细胞的体外杀伤作用。方法分别用重组腺病毒Ad-KDRp-CDglyTK、Ad-CMV-CD、Ad-CMV-TK、Ad-CMV-CDglyTK转染胃癌SCG7901细胞,给予前药5-FC、GCV,测定SCG7901细胞的集落形成、细胞存活率及该治疗系统的旁观者效应。流式细胞仪检测重组腺病毒Ad-KDRp-CDglyTK和Ad-CMV-CDglyTK分别对SCG7901细胞的细胞周期的变化及凋亡率。结果与Ad-CMV-CD、Ad-CMV-TK相比,Ad-KDRp-CDglyTK系统对SCG7901细胞的集落形成、细胞生长均具有更强的抑制作用,并可见较明显的旁观者效应。而与CMV启动子的双自杀基因相比,其作用差异无统计学意义。感染SCG7901细胞的两种腺病毒各自的实验组和对照组之间的细胞周期和凋亡率差异有统计学意义。结论 KDR启动子驱动的双自杀基因治疗系统对胃癌SCG7901细胞有确切的杀伤作用,其作用强度与强启动子CMV驱动的双自杀基因治疗系统相似。【Objective】 To evaluate killing effect of adenovirus containing cytosine deaminase and thymine kinase gene driven by vascular endothelial growth factor promoter(Ad-KDRp-CdglyTK) on stomach cancer cells in vitro.【Methods】 Ads of Ad-KDRp-CdglyTK,Ad-CMV-CD,Ad-CMV-TK and Ad-CMV-CDglyTK were transinfected into SCG7901 stomach tumor cells,followed by prodrugs(5-FC and GCV),respectively. Plating efficiency and MTT method were used to examine inhibition effect and bystander effect of the various Ads on SCG7901 cells.It were monitored that apoptosis rate and cell cycle of SCG7901 cells transfected by Ad-KDRp-CDglyTK and Ad-CMV-CDglyTK respectively with the methods of flow cytometry.【Results】 Compared with two single-suicide-systems,Ad-KDRp-CDglyTK system showed a more remarkable inhibiting effect on plating efficiency and growth of the tumor cells,and the bystander effect of the fusion suicide gene was much stranger than that of single suicide gene in vitro.But the SCG7901 cells infected with Ad-KDRpCDglyTK exhibited the similar sensitivity to prodrugs to the SCG7901 cells infected with Ad-CMV-CDglyTK. It showed that there were significance diffrernce in apoptosis rate of SCG7901 cells transfected by Ad-KDRpCDglyTK and Ad-CMV-CDglyTK respectively between control group and treatment group.【Conclusion】 AdKDRp-CDglyTK has effective antitumor effect on stomach tumor cells in vitro,and its effect wasn't significant different from that of Ad-CMV-CDglyTK.
