摘　　要：目的观察anti-miR-155反义寡核苷酸(AMOs)对人胰腺癌细胞株SW1990增殖和凋亡的影响。方法将SW1990细胞分为空白组、阴性对照组和AMOs处理组。阴性对照组将阴性对照链转染入SW1990细胞株中,AMOs处理组采用AMOs抑制SW1990细胞内miR-155的活性;CCK-8法测定细胞增殖抑制率,流式细胞术测定细胞周期及凋亡。结果与空白组及阴性对照组相比,AMOs显著抑制SW1990细胞的增殖(P<0.05),100nmol/L AMOs的抑制率达36.51%,使G0/G1细胞比例显著增加,G2/M期细胞比例显著减少(P<0.05),AMOs处理过的SW1990细胞早期凋亡率明显增高(P<0.05)。结论 AMOs通过抑制SW1990细胞内高水平表达miR-155的活性,显著抑制SW1990细胞的增殖。Objective To observe the effects of antisense-microRNA-155（AMOs） on apoptosis and proliferation of human pancreatic cancer cell line SW1990. Methods SW1990 cells were divided into three groups of BC（blank control）, NC（negative control） and AMOs（treated with AMOs）. Group NC was transfected negative control chain and the activity of miR-155 in the SW1990 ceils of group AMOs was inhibited by AMOs. The inhibition of cell proliferation was detected by CCK-8 and cell cycle and apoptosis were measured by flow cytometry. Results Compared with group BC and NC,the proliferation rate of SW1990 cells was obviously inhibited, the proportion of G0/Ga cells was increased significantly, and the proportion of Gz/M cells was significantly reduced in group AMOs（P〈0. 05）. The early apoptosis rate of SW1990 cells treated with AMOs was increased significantly（P〈0. 05）. Conclusion The proliferation of SW1990 ceils is significantly inhibited by AMOs through inhibiting the activity of miR-155 in SW1990 ceils.
