摘 要:目的探讨二维凝胶电泳和质谱分析在脑组织分析的应用价值。方法采用大脑中动脉阻塞建立大鼠脑梗死模型。采用二维凝胶电泳对正常大鼠脑组织和脑梗死后大鼠脑组织进行蛋白分离和图像分析;分离蛋白后进行质谱分析及数据库搜索,从而鉴定部分蛋白质斑点。结果筛选出5种差异性蛋白斑点,并通过数据库比对分析各自的功能。结论二维电泳凝胶图谱并作质谱分析可为研究脑梗死发病过程中蛋白质表达改变提供新的方法及途径。Objective To investigate the value of analyzing brain tissues with two-demensional electrophoresis and mass spectrometry in rats with cerebral infarction. Methods After establishment of rat model with cerebral infarction by ligation of the meddle cerebral artery,cerebral proteines from normal and infarcted brain tissues were separated by two-dimensional gel electrophoresis and irnage i analysis was earried out with PDQuest software. Protein spots were identified by the protein mass spectrometry and database searching. Results Five protein spots were identified successfully by mass spectrometry. Their function was confirmed by database searching. Conclusion The two-demensional electrophoresis and mass spectrometry can provide a new approach for further study on the changes of protein during ischemic cerebral infarction.
