摘  要:目的探讨普洱茶(熟茶)、铁观音、红茶对高脂饮食诱发的大鼠非酒精性脂肪肝中胰岛素样生长因子-I(insulin-like growth factor I,IGF-I)表达的影响。方法将Wistar大鼠随机分为正常对照组、高脂模型组、药物组及低、中、高剂量普洱茶(熟茶)、铁观音、红茶组(0.5,1.0,2.0 g/kg.bw)。试验大鼠灌胃授试茶样水浸提物35 d后采血处死,分离血清测定总胆固醇(TC)、甘油三酯(TG)、丙氨酸氨基转氨酶(ALT)、天冬氨酸氨基转氨酶(AST)的活性;用RT-PCR法检测试验大鼠肝脏细胞中IGF-I基因的表达量。结果与高脂模型组相比,药物对照组,中、高剂量普洱茶组,低、中剂量铁观音组,低、中剂量红茶组TC水平显著降低(P<0.05,P<0.01);药物对照组,低、中、高剂量普洱茶组,低、中剂量铁观音组,低、中、高剂量红茶组TG水平显著降低(P<0.05,P<0.01);低、中、高剂量普洱茶组,低、中剂量铁观音组和高剂量红茶组AST水平显著降低(P<0.05,P<0.01);低、中、高剂量普洱茶组和低、高剂量红茶组ALT水平显著降低(P<0.05,P<0.01);高剂量普洱茶、中剂量铁观音组IGF-I基因表达较高。结论普洱茶(熟茶)、铁观音、红茶对IGF-I在非酒精性脂肪肝中的基因表达具有不同程度的积极作用,普洱茶(熟茶)的作用更好。Objective To approach the effects of the fermented Pu-erh tea, Tieguanyin tea and Black tea on the expression of hepatocyte IGF-I in rats with non-alcoholic steatosis induced by high fat diet. Methods Wistar rats were randomly divided into control group, hyperlipidemia model group, medicinal group, and low, medium and high dose groups(0.5, 1.0, 2.0 g/kg.bw) of fermented Pu-erh tea, Tieguanyin tea and Black tea individually. All rats were sacrificed after 35 days' intragastric administration with the aqueous extracts of tested teas. The serum levels of TC and TG and the activities of ALT and AST were determined. The expression quantity of insulin-like growth factor I (IGF-I) in hepatocyte was detected by RT-PCR method. Results Compared to hyperlipidemia model group, the serum levels of TC were significantly decreased(P〈0.05, P〈0.01)in medicinal group, medium and high dose groups of fermented Pu-erh tea, low and medium dose groups of Tieguanyin tea, and as well as low and medium dose groups of Black tea; the serum levels of TG were significantly decreased(P〈0.05, P〈0.01)in medicinal group, low, medium and high dose groups of fermented Pu-erh tea, low and medium dose groups of Tieguanyin tea, and as well as low, medium and high dose groups of Black tea. Compared to hyperlipidemia model group, the serum levels of AST were significantly decreased(P〈0.05, P〈0.01) in low, medium and high dose groups of fermented Pu-erh tea, low and medium dose groups of Tieguanyin tea, and as well as high dose group of Black tea; the serum levels of ALT were significantly decreased(P〈0.05, P〈0.01) in low, medium and high dose groups of fermented Pu-erh tea, and as well as low and high dose groups of Black tea; the expression of IGF-I were increased in high does group of fermented Pu-erh tea and medium does group of Tieguanyin tea. Conclusion Fermented Pu-erh tea, Tieguanyin tea and Blacktea can have different degrees of positive effects on the expression of IGF-I in rat non-alcoholic steatosis m
