摘 要:高质量的RNA是进行基因克隆、Northern杂交分析等研究的必要前提。针对黄金梨果实中多酚、多糖等次生代谢产物含量较高的特点,综合前人的多种方法,选取其中3种方法进行比较,结果表明:改良3%CTAB法和TIANGEN RNAplant plus Reagent法均能提取出黄金梨果实中的总RNA,但后者更适于黄金梨果实总RNA的提取,得到的RNA经电泳检测后,条带无降解且亮度较好,能基本满足后续试验的要求。Isolation of high-quality RNA is the necessary premise of gene cloning and Northern blot.The Whangkeumbae Pear has the characteristic of the high level of polyphenol,polysaccharides and other secondary metabolities.Numerous methods of the predecessors have been taken into consideration,and three of them were chosen to be utilised to compare.The result shows that:modified 3% CTAB method and TIANGEN RNAplant plus Reagent were suitable for total RNA extraction from Whangkeumbae Pear,but TIANGEN RNAplant plus Reagent is more suitable.The electrophoresis of RNA was showed less degradation and higher purity.
