摘 要:目的检测南瓜多糖中特征官能团含量,考查特征官能团与其抗蛋白质非酶糖基化的关系。方法水提醇沉法提取南瓜粗多糖并分别以20%、40%和60%的乙醇分级醇沉为3种不同分子量的南瓜多糖,依次命名为PPⅠ、PPⅡ、PPⅢ。以苯酚硫酸法检测3种多糖中的糖含量;乙酰丙酮分光光度法检测氨基糖含量;硫酸咔唑法检测糖醛酸含量;硫酸钡比浊法检测硫酸基含量。根据线性相关分析法研究这些特征基团的含量与它们抗蛋白质非酶糖基化活性之间的关系。结果 PPⅠ、PPⅡ、PPⅢ的糖含量分别为92.62%、85.66%和79.60%;氨基糖含量分别为1.21%、1.95%和6.99%;糖醛酸含量分别为6.35%、4.64%和3.89%;3种多糖中均未检测到硫酸基的存在。南瓜多糖的抗糖基化活性与南瓜多糖的氨基糖含量呈正相关;与南瓜多糖的分子量及糖醛酸含量呈负相关。结论南瓜多糖中含有氨基糖和糖醛酸特征结构;南瓜多糖抗蛋白质非酶糖基化活性与其分子量、氨基糖及糖醛酸含量有关。Objective To investigate characteristic functional groups content of pumpkin poly- saccharides and relationship between the characteristic functional groups and anti-glycation ac- tivity. Methods Pumpkin polysaccharides were isolated and purified by the method of water extraction and ethanol precipitation. Then total pumpkin polysaccharides were divided into three parts with different molecular weight through different concentration of ethanol with the concentration of 20%, 40% and 60%, respectively. They were named after PP I , PP Ⅱ and PPⅢ. Sugar content,amino sugar content, uronic acid content and sulfuric acid group content of three PPs were determined by the way of phonel-sulfate method, acetylacetone coloration, sulfate-carbazole method and barium sulfate turbidimetric method, respectively. The relation- ship between their characteristic functional groups and anti-glycation activity was also dis- cussed. Results Three different molecular weight PPs were obtained. Detection of PP I , PP Ⅱ and PPⅢ showed that: sugar content were 92.62%, gS. 66% and 79.60% ; amino sugar content were 1.21% , 1.95% and 6.99% ; uronic acid content were 6.35% , 4.64% and3.89%, and there was no sulfuric acid existing in PPs. According to the linear correlation analysis, the anti-glycation activity of pumpkin polysaccharides were positively correlated to their amino sugar content and negatively correlated to their molecular weight and uronic acid content. Conclusion Pumpkin polysaccharide contains amino sugar and uronic acid, the anti- glycation activity of pumpkin polysaccharides are correlated to their molecular weight, amino sugar and uronic acid content.
