摘  要:采用响应面法对重组大肠杆菌谷氨酸脱羧酶转化谷氨酸生成γ-氨基丁酸的条件进行优化。首先通过单因素实验得出酶转化的初步条件,再经过Plackett-Burman设计法筛选出主要影响因素,并利用响应面分析法对各主要因素进行优化,最终确定的最佳条件为:底物浓度0.290 mol/L,pH值4.99,转化温度52.10℃,转化时间60 min 22s。采用优化后的转化条件进行转化酶活达到了1461.84μmol/(5 mL·h),比优化前提高了19.11%。The response surface methodology was used to optimize the conversion conditions of γ-aminobutyric acid ( GA-BA) for recombinant E. coli glutamate decarboxylase (GAD). Based on the single-factor test, the main factors were found by Plackett-Burman design. Then the response surface methodology was used to optimize the main factors. The opti-mized conditions were as follows : 0.290 mol/L substrate concentration, pH 4.99, 52.10 ℃ transformation temperature and 60 rain 22 s transformation time. Under the optimum conditions the activity reached 1461.84μmol/(5mL·h), which was increased by 19.11% than that of before.
