摘 要:目的应用慢病毒介导的RNA干扰技术,探讨RNA干扰沉默fascin基因的表达对人胆管癌细胞QBC939体外侵袭力的影响。方法采用常规方法培养QBC939细胞,将其分为三组,实验组和对照组分别转染fascin shRNA慢病毒载体和shRNA阴性对照慢病毒载体,空白组不做处理。Western blot检测各组fascinmRNA和蛋白的表达情况;通过Transwell侵袭试验检测各组细胞在体外的侵袭能力。结果 Western blot检测发现,实验组fascin基因的表达明显低于空白组及对照组,差异有显著性(P<0.05);Transwell侵袭试验检测发现实验组侵袭能力明显低于其余两组,差异有显著性(P<0.05))。结论慢病毒介导的shRNA能有效地沉默Fascin基因在胆管癌QBC939中的表达,能有效抑制胆管癌细胞的侵袭能力。【Objective】 To study the effect of lentivirus-mediated RNA interference(RNAi) of fascin on the metastatic potential of human Cholangiocarcinoma cell line QBC939 in vitro.【Methods】 QBC939 cells was conventionally cultured.A lentiviral vector for RNAi targeting the coding region of human fascin and another control vector containing a nonsense sequence were constructed.The vectors were transfected into QBC939 cells,and fascin expression in the cells was detected by Western blotting.The invasive ability of the transfected QBC939 cells was evaluated using Transwell.【Results】 Compared with the control vector,fascin-RNAi-LV resulted in obviously reduced expression of fascin protein,lowered invasive ability of QBC939 cells(P &lt;0.05).【Conclusion】 Lentivirus-mediated nestin RNAi can specifically inhibit Fascin expression to cause decreased cell invasiveness of human cholangiocarcinoma cell line QBC939.
