摘  要:采用2-油酸-1-棕榈酸磷脂酰胆碱为表面活性剂、低级醇为助表面活性剂、异辛烷为油相、蒸馏水为水相,以伪三元相图微乳面积大小为指标,考察磷脂酰胆碱微乳体系。结果表明,磷脂/正丙醇/异辛烷/水W/O微乳体系、磷脂/正丁醇/异辛烷/水W/O微乳体系以及磷脂/正丙醇/异辛烷/水O/W微乳体系均为稳定的磷脂酰胆碱微乳体系。3种微乳体系和脂质体分别与鸭肉肌内磷脂酶振荡反应10 h后,采用TLC联用Quantity One软件鉴定反应产物中的脂肪酸,以脂肪酸斑点分析的Trace Qty值为指标,最终确定磷脂酰胆碱反应体系。因此,以脂质体作为反应体系所产生的脂肪酸最多,即脂质体为磷脂酰胆碱最佳反应体系。为进一步研究磷脂酰胆碱的酶解机制和磷脂酶的分离纯化提供技术方法。The desired microemulsions were prepared by mixing isooctane as organic phase,distilled water as water phase,2-oleoyl-1-palmitoyl-sn-glycero-3-phosphocholine as solvent,low carbon alcohols as co-solvents.The size of microemulsion region of the pseudo-ternary phase diagram was used as the stable index of microemulsion to study,phosphatidylcholine microemulsion systems.The results showed that phosphatidylcholine/n-propanol/ isooctane W/O microemulsion,phosphatidylcholine/n-butanol/ isooctane W/O microemulsion,phosphatidylcholine/n-propanol/ isooctane O/W microemulsion were all stable systems.Three microemulsion systems and liposome were in touch with the intramuscular phospholipase extracted from duck muscle respectively,and the reaction time was ten hours.Fatty acids contents as the main hydrolysates were identified with Tract Qty by TLC and Quantity One to estimate the optimal phosphatidylcholine reaction system.It showed that the generation of fatty acids was more frequently in liposome as reaction system.Finally,liposome was determined as phosphatidylcholine reaction system to set up a technical method for purification and characterization of phospholipase and the mechanism of phospholipid hydrolysis.
