摘 要:目的探讨茶多酚(TP)对草甘膦致小鼠睾丸支持细胞(SC)损伤的拮抗作用。方法原代培养小鼠睾丸支持细胞,采用FasL免疫细胞化学法鉴定支持细胞。建立草甘膦诱导支持细胞损伤模型,将细胞随机分为正常对照组、草甘膦损伤组(90btg/m1)、TP拮抗组(经10、20、40、80、160μg/mlTP预处理12h后,再用草甘膦作用细胞24h)。四噻唑蓝(MTT)法检测细胞存活率,倒置相差显微镜观察细胞形态学改变,原位末端标i5(TUNEL)法原位检测细胞凋亡,Hoechst33342荧光染色法观察细胞核形态改变,并检测细胞氧化损伤指标超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSH—Px)、丙二醛(MDA)的变化。结果10~80μg/mlTP可拮抗草甘膦诱导的支持细胞损伤,提高支持细胞活力及SOD、GSH—Px活性,降低细胞凋亡指数及MDA含量,改善细胞萎缩变形、染色质浓集现象;160μg/ml时不能减轻草甘膦造成的细胞损伤。结论TP在一定浓度范围内对草甘膦诱导的支持细胞损伤具有拮抗作用。Objectives To observe the protective eltect ot tea polyplaenols (IP) on me oamage lnouceo oy glypnosae in the sertoli cells (SC). Methods SC from weaning mice were obtained and cultured. The SC were identified by immuno-cytochemical reaction. The damage model of SC was induced by glyphosate. The cells were divided randomly into control group, glyphosate model group and TP protective group (the SC pretreated with 10, 20, 40, 80, 160 ktg/ml TP for 12 h, and then glyphosate added to SC for 24 h respectively) . The cell viability was measured by methylthiazolyl blue tetrazolium (MTT), and the morphological observation of SC was performed under inverted phase-contrast microscope. The cell apoptosis was observed by TdT-mediated dUTP nick end labeling (TUNEL), and their nuclei were observed with Hoechst33342 fluorescent staining. Super oxide dismutase (SOD), GSH-Px, MDA were measured. Results TP played the protective role against SC damage induced by glyphosate between the concentration from 10 to 80 μg/ml. TP increased the cell viabilities, SOD and GSH-Px activities. Meanwhile the cell apoptosis index, the level of MDA and chromatin concentration decreased. The protective effect of TP on damage of cells could not be observed at 160μg/ml. Conclusion There is protective effect for TP on the damage of SC induced by glyphosate at appropriate concentration.
