摘 要:目的:建立保健食品中水飞蓟宾的高效液相色谱测定方法。方法:采用WATERS Xterra RP18(4.6×250 mm,5μm)色谱柱分离;流动相为甲醇:0.5%冰醋酸=48:52(V/V);流速1.00 ml/min;2487紫外检测器287 nm测定。结果:水飞蓟宾在1.0μg/ml~50μg/ml时,浓度与峰面积呈良好的线形关系(r=0.9998),检出限为0.20μg/ml,加标回收率在94.3%~98.0%之间,相对标准偏差RSD在2.40%~2.66%之间。结论:该方法简便、快速、准确,适用于保健食品中水飞蓟宾含量的测定。Objective: To establish a HPLC method for the determination of silybin in health food. Methods : The separation was carried out on a Xterra RP18 (4.6 × 250mm,5 um) column with 2487 UV detector at 287nm , the mobile phase were methanol and 0.5% acetic acid(48:52 v/v) with the flow rate at 1.00 ml/min. Results:When the silybin was at 1.0 ug/ml - 50 ug/ml, the perfect linearity between concentration and peak area ratio was ob- tained(r = 0. 9998 ). The detection limit of glutathione was 0.20 ug/ml, the recovery was 94.3% - 98: 0% by adding standard to sample solution and the RSD obtained for the within - run precision was between 2.40% - 2. 66%. Conclusion:This method is simple,rapid and accurate, so it can be used for the determination of silybin in health food.
