摘 要:从造纸厂污泥中获得一株产高温纤维素酶的真菌XM5,经形态学和ITS序列分析,鉴定其为土曲霉(Aspergillus terreus).在稻草液体发酵培养基中,土曲霉XM5所产纤维素酶的合成模式为同步合成型.酶学性质研究表明,该纤维素酶的最适作用温度是65℃,在80℃下保温2 h活力残留40%以上,最适作用pH为4.0,在pH4.0~7.0内较稳定,实验结果表明,该菌株所产纤维素酶具有较好的温度稳定性和pH稳定性.A wild strain with high temperature tolerance cellulase was isolated from the soil samples near the paper mill.The strain was identified and named as Aspergillus terreus XM5 by its morphological analysis and ITS rDNA sequence.The pattern of cellulase synthesis was associated with the growth of Aspergillus terreus XM5 in submerged fermentation with rice straw as the sole carbon source.The optimum temperature for the cellulase activity was at 65 ℃ and the cellulase maintained more than 40% of its maximum activity after incubation for 2 h at 80 ℃.The optimum pH for the cellulase activity was at 4.0;and the cellulase retained stability in the pH range of 4.0 ~ 7.0 after incubation for 3 h.The results showed that the cellulase from the strain has high pH and temperature stability.
