摘  要:从湖北地区棉花地采样,筛选分离出一株纤维素分解菌株W2,根据形态及18S rDNA序列分析,初步鉴定为木霉属。以玉米芯为发酵原料,对W2固态发酵(SSF)产羧甲基纤维素酶(CMCase)的条件进行优化。选取发酵时间、温度、基质含水量和初始pH为自变量,以CMCase活力为响应值,采用Box-Benhnken中心组合设计和响应面分析法,研究了各变量及其交互作用对CMcase的影响,建立了二次多项式回归方程及响应面图。结合单因素试验,确定了适宜的发酵条件为:温度25℃,基质含水量63%,初始pH6.0,发酵65h,CMCase活力达到最大。最后,在适宜的固态发酵曲中,加入活化酵母细胞,采用改良Conway法测定发酵曲中酒精的含量,达0.38mg/mL。A strain W2 was separated from the rotted cotton of Hubei province. According to morphology and 18S rDNA sequences analysis, strain W2 belongs to Trichoderma sp. Corn cob was used as substate to optimize solid-state fermentation (SSF) conditions for CMCase production. Four parameters including fermention time, temperature, initial moisture content and pH were optimized and the CMCase activity was acted as response value. The interaction of the respective factors and their influences on CMCase were studied by Box-Benhnken central composite design and response surface methodology, the quadratic equation and response figures were established. The optimal fermentation conditions were obtained based on single-factor tests. Under culture temperature of 25℃, initial moisture content of 63% and pH 6.0, CMCase achieved its high value after 65h fermentation. At last, synchronous fermentation was adopted to produce alcohol, and the alcohol content reached 0.38 mg/mL determined by improved Conway method.
