支原体液体培养试剂冻干质控品制备研究

摘　　要：目的:研究支原体液体培养试剂菌种质控品合适的冻干保护剂。方法:把高浓度的支原体菌种分别按1:15.7和1:332的比例稀释到2%脱脂奶粉、2%海藻糖、2%胰酪胨、2%甘露醇、2%甘油和SP-4培养基中,以1:15.7比例冻干质控品复溶液的比例代表≧104CCU/ml,以1:332冻干质控品复溶液的比例代表<104CCU/ml,并以此质控2种有代表性的支原体液体培养试剂。结果:只有SP-4培养基能够满足性能质控的要求。结论:可以以SP-4培养基作为冻干保护剂制备支原体液体培养试剂菌种质控品。Objective： To prepare the freeze - drying quality control specimens of Mycoplasma in liquid culture. Methods ： In ratio of 1 ： 15.7 and 1：332, high - concentration Mycoplasma stock cultures were diluted with 2% defatted milk powder, 2% trehalose, 2% trypticase, 2% mannitol, 2% glycerol and SP - 4 medium, respectively. Two kinds of representative Mycoplasma liquid culture reagent as a model, quality control were performed with re - dissolved solution of lyophilizedquality control specimens inl ：15.7 （ 〈 104 CCU/ml） and 1：332 （ 〈 104 CCU/ml）. Results： Only SP - 4 medium can meet the performance requirements of quality control. Conclusion ： SP - 4 media could be used as cryoprotectant for the preparation of quality control specimens of Mycoplasma in liquid culture.
