摘 要:目的通过测定血管瘤内皮细胞表达Cyclin D1、p16、CDK4的水平,探讨儿童血管瘤的增生、退化机制。方法将31例儿童血管瘤蜡块及6例儿童正常皮肤组织蜡块分为血管瘤增生组、退化组、血管畸形组及对照组。对4个对比组血管内皮细胞Cyclin D1、p16、CDK4的表达及其光密度值和阳性面积率进行统计学分析。结果 Cyclin D1、p16、CDK4在血管瘤增生组、退化组呈高表达,而在血管畸形组及对照组低表达,各组间蛋白表达的光密度值和阳性面积率进行比较,增生组和退化组明显高于血管畸形组及对照组,差异有统计学意义(P<0.01),且Cyclin D1和p16表达的光密度值呈正相关(r=0.68,P=0.00)。结论 p16突变增多对CDK4的活性抑制作用减弱。血管内皮细胞Cyclin D1和CDK4增多,可能是真性血管瘤发生和增殖的一个重要因素。【Objective】 To investigate the proliferation and devolution mechanisms of hemangioma and vascular malformation in children through quantitating the expression of Cyclin D1,p16 and CDK4 in endothelial cell of hemangioma.【Methods】 Thirty one hemangioma and six normal skins of paraffin blocks in children were collected and divided samples into proliferative stage,involuting stage,vascular malformation and contrast groups.The values of light densities and positive area rates which Cyclin D1,p16 and CDK4 expressed on endothelial cell in different groups were analyzed with patho-image analytical technique statistically.【Results】 There were homo-express of Cyclin D1,p16 and CDK4 in proliferative and involuting groups and hypo-express in vascular malformation and contrast groups.The values of optical density and positive area rate which Cyclin D1,p16 and CDK4 expressed were higher obviously in proliferative and degradative groups than that in groups of vascular malformation and control compared among groups with statistical significance(P 0.01).The linear correlation is positive between p16 and Cyclin D1(r =0.68,P =0.00).【Conclusion】 The endothelial cells of hemangioma in children have growth vigor because of mutational p16 cannot inhibit the activity of CDK4 and the Cyclin D1 and CDK4 promote proliferation in endothelial cells.
