基质金属蛋白酶9基因-1562C／T多态性与缺血性卒中风险：汇总分析

摘　　要：目的探讨基质金属蛋白酶9（matrix metalloproteinase9，MMP-9）基因-1562C／T多态性与缺血性卒中风险的相关性。方法检索PubMed、Embase、MEDLINE、webofscience以及CBMdisc和CNKI数据库，收集2013年5月之前公开发表的所有相关研究。提取数据后，应用Review Manager 5．0和Stata11．0软件包对符合要求的研究进行汇总分析。合并效应量并计算优势比（oddsratio，OR）和95％可信区间（confidenceinterval，CI）。结果共纳入6项研究，包括1115例患者和979例对照者。合并分析后得出的OR（95％CI）分别为：共显性遗传模式C／T对C／C：1．10（0．71～1．71），T/T对C／C：1．78（0．89～3．57）；显性遗传模式C／T＋T/T对C／C：1．13（0．76～1．67）；隐性遗传模式T/T对C／T＋C／C：1．79（0．90～3．57），提示MMP一9基因-1562C／T多态性与缺血性卒中风险无显著相关性。将来自中国人群的5项研究进行的分析显示，上述4种遗传模式的OR（95％CI）分别为1．24（0．76～2．04）、1．43（0．52～3．92）、1．25（0．80～1．97）和1．33（0．49～3．62），同样无显著性统计学意义。结论根据现有文献分析，MMP-9基因-1562C／T多态性与缺血性卒中风险无显著相关性。Objective To investigate the correlation between the - 1562 C/T polymorphism of matrix metalloproteinase-9 （MMP-9） and the risk of ischemic stroke. Methods All the relevant literatures published were searched from PubMed, Embase, MEDL1NE, Web of Science as well as CBMdisc and CNKI before May 2013. After extracting data, a meta-analysis of the studies meeting the requirements was conducted by using Review Manager Version 5.0 and Stata 11.0 software. The effect size was pooled and odds ratio （OR） and 95% confidence interval （CI） were calculated. Results Six studies were enrolled, including 1 115 patients and 979 controls. The ORs after pooled analysis （95% Cls） were C/T vs. C/C： 1.10 （0. 71 - 1.71） and T/T vs. C/C： 1.78 （0. 89 - 3.57） of the codominant effetic model, respectively; C/T ＋ T/T vs. C/C： 1.13 （0. 76 - 1.67） of the dominant genetic model; T/T vs. C/T ＋ C/C： 1.79 （0. 90 - 3.57） of the recessive genetic model, which sugggsted that there was no sigtificant correlation between the - 1562 C/T polymorphism of MMP-9 gene and the risk of ischemic stroke. The analysis of 5 studies in Chinese populations showed that the ORs （95% Cls） of the above 4 genetic models were 1.24 （0. 76 - 2. 04）, 1.43 （0. 52 - 3.92）, 1.25 （0. 80 - 1.97）, and 1.33 （0. 49 - 3.62）, respectively. There were also no significant differences. Conclusions According to the analysis of the available literatures, there was no significant correlation between the - 1562 C/T polymorphism of MMP-9 gene and the risk of ischemic stroke.
