摘 要:目的针对老鹰茶(豹皮樟)野生自然繁育难,资源更新周期长的情况。初步探讨豹皮樟的组织培养技术。方法以老鹰茶的叶、叶柄,茎段、芽孢、种子为外植体,在MS+BA2.0 mg·L-1+NAA0.2 mg·L-1的培养基培养,观察愈伤组织状况;在其组织培养工作中污染问题较严重,该实验中采用了3种消毒方法:(1)75%酒精消毒30 s→10%次氯酸钠浸泡10 min→无菌水洗3次;(2)0.1%升汞液灭8 min→10%次氯酸钠浸泡10 min→无菌水洗3次;(3)75%酒精消毒30 s→0.1%升汞液灭菌8 min→无菌水洗3次。结果早春和秋后芽孢为最优外植体,污染率相对较低,愈伤组织诱导率达36.4%,表面消毒以第3种方法效果最好。结论由早春、秋后的芽孢为外植体,初步得到了老鹰茶组织培养的无菌材料,为进一步的研究提供了基础。Objective To explored tissue culture to solve the propagate problem in the production with Laoying tea(Litsea coreana).Methods The leaves,stems,petiole,bud and seeds were used as explants to induce callhs in MS + BA 2.0 mg.L-1 + NAA 0.2 mg.L-1.There were three variants of surface sterilization.The first explant was soaked in 75% C2H5OH for 30 s-10% NaOCl for 10 min-3 times sterile water,the second was soaked in 0.1% HgCl2 for 8 min-10% NaOCl for 10 min-3 times sterile water,the third was soaked in 75% C2H5OH for 30 s-0.1% HgCl2for 8 min-3 times sterile water.Results The best explants were bud in early spring and autumn.The callus inducing ratio was as high as 36.4% and the pollution was low.The most suitable variant for surface sterilization of explants was the third one.Conclusion Obtaining aseptic materials from buds in early spring and autumn would provide basis for further research.
