摘  要:应用高效液相色谱检测饲料中丙二醛的含量,建立一种简单、快速、定量评估饲料油脂酸败程度的方法。选取鸡、猪、宠物和水产等不同饲料样本,经三氯乙酸提取,TBA衍生化,以乙腈:磷酸二氢钾(50mmol/L)(V/V=18/82)为流动相、C18柱分离,荧光检测器检测。结果表明:在0.02~0.20μg/mL,丙二醛的峰面积与其质量浓度间有良好的线性关系,相关系数为0.9994,定量限(以信噪比为10计)为0.05mg/kg。在鸡、猪、宠物等7种饲料样本基质中添加1、2、5μg/kg丙二醛,其平均回收率为85%~96%,相对偏差均小于5%,满足饲料中丙二醛检测的要求。Abstract.A simple, rapid and quantitative method to evaluate the oil rancidity of feed was established through determining the malondialdehyde content in' feed by high performance liquid chromatography(HPLC). The samples from feeds of poultry,pigs,pets and fishes were extracted with trichloroacetic acid and derived by thiobarbituric acid(TBA), then analyzed by separation on a C18 chromatographic column with an acetonitrile and KH2 PO3 (V/V = 18/82)solution as a mobile phase, and detected with a fluorescence detector. The results showed that there was a good linear relationship between the peak area of malondialdehyde and the mass concentration within 0.02 -0.20 mg/L, the correlation coefficient was 0. 9994 and the limit of quantification(S/N = 10)was 0.05 mg/kg. When 1,2 or 5 μg/kg malondialdehyde was added to 7 feed samples, the average recovery rates were 85 % - 96%, and all the relative standard deviations(n = 10)were less than 5%,meeting the requirement for deter- mining malondialdehyde content in feed.
