摘  要:非极性代谢产物的分离和含量分析是植物代谢组学的重要组成部分。在非极性代谢产物的分离过程中,样品提取方法对于化合物的分离种类和分离量具有十分重要的影响。本文以草地早熟禾品种‘午夜’的叶片为材料,采用甲醇-氯仿提取法将非极性代谢产物从叶片中分离出来,然后再采用酸或碱水解法进行水解和甲酯化反应。经过酸水解后30种化合物被分离出来,采用碱水解的方法后,36种化合物被分离出来。而且采用碱水解后,大部分化合物的分离量显著高于酸水解。这种甲醇-氯仿提取结合碱水解的方法可以作为简单、高效的禾本科植物样品非极性化合物气相色谱-质谱前处理方法。The separation and content analysis of lipophilic metabolites are major components of plant metabolics. During the separation process of lipophilic metabolites, extraction methods play important roles to the kinds and contents of separated metabolites. Lipophilic metabolites were separated with methanol- chloroform method from leaves of Kentucky bluegrass (Poa pratensis ' Midnight) and then hydrolyzed and methylesterified under the condition of acid or alkaline catalysis. After hydrolysis catalyzed by acid and alkaline,30 and 36 kinds of lipophilic compounds were separated respectively. The contents of most of separated compounds by alkaline catalyzed hydrolysis were significantly higher than by acid catalyzed hydrolysis. Methanol-chloroform extraction combined with alkaline catalyzed hydrolysis can also be used as a simple, efficient preliminary treatment method for lipophilic compounds with gas chromatography-mass grass family species in extracting and separating of spectrometry.
