摘 要:【目的】 探讨咖啡因对平阳霉素诱发人精子染色体畸变的影响。【方法】 人精子经平阳霉素(pinyangmycin ,PYM)处理与去透明带地鼠卵受精 ,受精 1h后加入咖啡因 (caffeine ,CAF)后处理 ,继而制备染色体进行核型分析。实验分为对照、CAF、PYM、PYM +CAF 4个组。【结果】 CAF组、PYM组和PYM +CAF组染色体结构畸变精子率依次为 2 7%、40 %、76 % ;断裂均数依次为 0 72、1 92、4 36上述各组参数均高于空白对照组(8 0 %、0 14) ,差异有非常显著意义 (P <0 0 1)。与CAF组和PYM组比较 ,PYM +CAF组诱发的染色体型和染色单体型畸变均明显增加 ,以染色单体型增加更为明显 (P <0 0 1)。【结论】 咖啡因可诱发人精子染色体畸变 。Objective To explore the effects of caffeine on human sperm chromosome aberrations induced by pinyangmycin. Methods Human spermatozoa,which was treated with pinyangmycin,were fertilized in vitro with zone free hamster ova for one hour,then the fertilized eggs were treated with caffeine.Chromosome aberrations were analyzed at the first cleavage metaphase of fertilized eggs.The experiments were divided into four groups:control,caffeine,pinyangmycin(PYM) and PYM +caffeine groups. Results Frequencies of chromosome aberrations in caffeine,PYM and PYM+caffeine groups were 27%,40% and 45%,and the means of breakage were 0 72,1 92 and 4 36 sequentially.All of above parameters were more higher than those in controls(8%,0 14)( P <0 01),Compared with those in caffeine and PYM groups,the increase in the frequencies of chromosome type and chromatid type aberrations were highly significant in PYM+caffeine groups( P <0 01).The frequencies of chromatid type aberrations were more higher than those of chromosome type. Conclusion Caffeine could induce chromosome aberrations in human spermatozoa and potentiate the effects of chromosome aberrations induced by PYM.
