摘 要:为获得接骨草叶中绿原酸的最佳提取工艺,比较了水煮法、乙醇浸提法、超声波辅助提取法和索氏提取法提取接骨草叶中绿原酸,用紫外分光光度法测定其含量。结果表明,从接骨草叶中提取绿原酸的最佳方法为超声辅助提取法。通过设计正交实验得到超声波提取法的最佳提取工艺是乙醇浓度为80%,固液比为1:45(g/mL),超声波辅助提取时间为45min,而各因素对提取绿原酸的影响依次为固液比〉乙醇浓度〉超声辅助提取时间。在最佳提取条件下,一年中6.11月间接骨草叶中绿原酸得率分别为1.852%、1.995%、1.944%、1.891%、1.803%和1.742%,故采收接骨草叶提取绿原酸的最佳月份是7月。The optimum extraction process explored through four different methods of of chlorogenic acid in the leaves poach method, ethanol extraction, of Sambucus chinensis Lindl.was utrasonic-assisted extraction, and soxhlet extraction. Then the content of chlorogenic acid was determined by ultraviolet spectrophotometry. The results showed that the optimal extraction method for chlorogenic acid extraction from the leaves of Sambucus chinensis was utrasonic-assisted extraction.The orthogonal design L9 (3^3) was employed to study the effects on chlorogenic acid leaching rate by investigating the concentration of ethanol, solid-liquid ratio, ultrasound time on the utrasonic-assiste extraction.The effects of factors on chlorogenic acid leaching rate were as follows:solidliquid ratio, ethanol concentration, ultrasonic extraction time. The optimal extraction conditions were 80% ethanol concentration,the ratio of 1:45 (g/mL), ultrasonic extraction time 45min. Under the optimal extraction condition, chlorogenic acid yield in June to November was 1.852%, 1.995 %, 1.944%, 1.891%, 1.803%, 1.742% respectively.The best harvest time of leaves for extraction chlorogenic acid was in July.
