摘 要:建立肌肉(猪肉、牛肉)及牛奶中6种玉米赤霉醇类化合物(α-玉米赤霉醇、β-玉米赤霉醇、α-玉米赤霉烯醇、β-玉米赤霉烯醇、玉米赤霉酮和玉米赤霉烯酮)残留量的免疫亲和柱净化-高效液相色谱同时检测方法。试样酶解后,肌肉样品经乙醚提取,牛奶样品经乙腈提取,免疫亲和柱净化,高效液相色谱仪测定。结果表明,6种目标物在0.01~0.2μg/mL范围内线性关系良好,相关系数(r)均大于0.999,检出限(S/N≥13)为1.0μg/kg,猪肉、牛肉、牛奶中的平均回收率分别在72.8%~88.4%、72.6%~92.4%、82.3%~102.3%之间,RSD均小于10.0%。该方法灵敏度高、重现性好,适用于猪肉、牛肉、牛奶等动物源性食品中痕量玉米赤霉醇类药物残留的测定。An immunoaffinity cleanup- high performance liquid chromatography (IAC- HPLC)method was established for the simultaneous determination of residues of six kinds of zeranols (co-zeranol, α-zeranol, β-zearalenol,α-zearalenol, zearalanone and zearalenone)in pork, beef and milk. Enzymolyzed samples were extracted with organic solvent (muscle by Ethyl ether, and milk by Acetonitrile), subsequently cleaned up on a immunoaffinity column,and determined by HPLC.Six kinds of zeranols had good linear relationships in the range of 0.01~0.2μg/mL, the correlation coefficient was higher than 0.999, the limit of detection was 1.01μg/kg for each zeranol(S/N≥13).Average recoveries of six target compounds in pork, beef and milk at different concentrations ranged was 72.8%~88.4% ,72.6%~92.4% and 82.3%~102.3%, relative standard deviation were less than 10.0%.The method could be used for determination of trace residues of six kinds of zeranols in foodstuffs of animal origin,and stable, reliable and accurate.
