摘  要:目的研究儿茶素单体表没食子儿茶素没食子酸酯(EGCG)和表儿茶素没食子酸酯(ECG)抗人肝癌细胞BEL-7402的作用。方法形态学观察EGCG、ECG作用后BEL-7402细胞的生长;MTT法检测EGCG、ECG对BEL-7402细胞生长的抑制;流式细胞术(FCM)检测EGCG、ECG诱导BEL-7402细胞凋亡水平;RT-PCR检测增殖相关信号分子Gli2和凋亡受体Fas的表达水平。结果 EGCG、ECG均能以时间依赖和浓度依赖的方式抑制BEL-7402细胞生长,并诱导细胞凋亡。EGCG对BEL-7402细胞的生长抑制和诱导凋亡作用均较ECG强。EGCG和ECG均能下调Gli2的表达,但对Fas表达无影响。结论EGCG抗人肝癌BEL-7402细胞作用较ECG强。Objective To investigate the anti-tumor activities of Epigallocatechin gallate(EGCG) and Epicatechin gallate(ECG) against human hepatocarcinoma BEL-7402 cells.Methods Morphology of BEL7402 cells treated with EGCG or ECG was observed under microscope.Cell proliferation was assayed by MTT and growth inhibitory rate was calculated.Apoptosis of BEL-7402 was detected by flow cytometry.The expression of proliferation-related Gli2 and apoptosis receptor Fas genes was assayed by RT-PCR.Results Both EGCG and ECG inhibited cell growth and induced apoptosis of BEL-7402 cells in a timedependent and concentration-dependent manner.EGCG is more effective on cell growth inhibition and apoptosis induction than ECG.Both EGCG and ECG decreased Gli2 expression,but not for Fas expression.Conclusion The anti-tumor activity of EGCG against BEL-7402 cells is stronger than that of ECG.
