结冷胶菌种的甘油保存方法研究

摘  要:本文通过对菌种保存方法的优化,找出了一种简单易操作的结冷胶菌种保藏方法。将甘油与生理盐水分别按不同的比例进行配比,作为结冷胶菌种保藏液,并将菌种保藏液与结冷胶菌悬液按不同的比例混合,做成甘油管,在温度-20℃下保存3个月后,各取一支进行试验。通过实验结果的对比,确定了结冷胶菌种保藏液中甘油与生理盐水的最佳比例为0.6:0.4(v/v),甘油保藏液与结冷胶菌种悬液的保藏优化比例为1:1(v/v)。用此比例将结冷胶菌种进行保藏,分别于6个月、1年、2年、3年后对菌种进行活化,检测菌种培养时间,菌体形态,发酵性能等,发现用此保藏方法将结冷胶菌种保藏3年后,除了菌种培养时间稍微延长之外,其它并无影响,从而验证了所得比例的可靠性。In this paper, a simple and easy-operating gellan gum culture collection method was found based on the optimization of species preservation method. Glycerol and saline were carried out in different proportions ratio as gellan gum liquid culture collections, and the culture Collection liquid with gellan gum bacterial suspension were mixed in different proportions, made of glycerin tubes, then stored at a temperature of -20 ℃for 3 months, and depicting a test. Comparing the results of the experiment, we got the optimal ratio of glycerol and saline was 0.6 : 0.4 ( v/v ) in culture collection liquid, the optimal ratio of glycerol preservation solution and gellan gum bacteria suspension was 1 : 1 ( v/v ) . Preserved gellan gum strains with the optimized ratios for six months, one year, two years and three years, and then activated the bacteria, detected the bacteria culture time, cell morphology, and fermentation performance, etc., we found that strain performances were almost no change after three years by using this method to preserved gellan gum culture collections.
