摘 要:角质酶(EC3.1.1.74)是水解酶,可以降解角质产生脂肪酸,属于α/β水解酶,也可以水解甘油三酯等化合物。角质酶的应用十分广泛,但菌株对角质酶的表达量非常有限,因此提高角质酶表达量的研究具有广泛的理论与实践意义。角质酶是一种诱导酶,角质的存在能激活产该酶病菌的角质酶基因,诱导其高效转录表达。本实验经过角质诱导及诱导强度的变化,真菌的角质酶基因转录量大大增加,而未经过角质诱导的真菌角质酶基因转录量非常少。本研究中诱导棉花枯萎病菌的产酶量最高可以达到29.46U/mL,比未诱导的提高至4倍。Cutinase (EC3.1.1.74) is a hydrolytic enzyme that can degrade cutin into fatty acids, belonging to the α/β hydrolase, and is able to hydrolyse triglycerides and other compounds. Cutinase can be used widely but the content of cutinase expressed by the strains is very low. Thus the studies focused on increasing the expression of cutinase have extensive theoretical and practical significance. Cutinase is an inducible enzyme, and the pres- ence of turin can activate the cutinase gene in the bacteria producing the enzyme and induce an efficient expression of the gene at transcription level. In this study, the transcription of cutinase gene in fungal was greatly increased as the induction with cutin and the increase of induction. The tran- scription level of ftmgal cutinase gene was very low without induction of cutin. In the present paper, the induction of cutin was carried out on bacte- ria Fusarium oxysporum and the highest enzyme activity could reach to 29.46U/ml, which was four times of the activity of F, oxysporum without induction.
