摘 要:目的 建立正常人垂体组织基因表达谱。方法 垂体cDNA文库构建、大规模表达序列标记 (EST)测序及生物信息学处理。结果 在所测的 72 2 2条可分析的EST中 ,2 949条 (4 1% )为已知基因 ,其中参与基因 /蛋白表达者最高 (2 3.2 % ) ,13 .7%的基因与内分泌相关 ,18.1%的基因参与信号转导。前 2位高表达的基因是促生长素和催乳素。垂体表达甘丙肽、可卡因和安非他明调节的转录肽 (CART)及黑色素浓集激素 (MCH)等与食欲调节有关的激素。还表达与免疫有关的细胞因子和受体。Pit 1、锌指蛋白 (ZNF) 74和PLAG1在垂体中是相对特异表达的。结论 正常垂体组织中参与信号转导的基因明显高于心脏和CD34 +造血前体细胞 ,而与脑、下丘脑、肾上腺组织相近。甘丙肽和CART可能是参与食欲调节的垂体激素。垂体和免疫系统之间存在广泛的相互作用。Pit 1、ZNF74和PLAG1可能是垂体特异表达的转录因子。Objective To establish the gene expression profile in normal human pituitary. Methods cDNA libraries of normal human pituitary were constructed. Large scale expressed sequence tags (ESTs) sequencing and bioinformatics were used. Results Among the 7222 ESTs obtained, 2949 (41%) were known genes. Within the identified genes, 23.2% were associated with gene/protein expression, 13.7% were endocrine related, 18.1% were related with cell signal transduction. Somatotropin and prolactin were the first two genes with the highest expression in normal pituitary. Galanin、 cocaine and amphetamine regulated transcript peptide (CART) and melanin concentrating hormone (MCH), etc., which were related to the regulation of appetite, were expressed in pituitary. Cytokines and receptors related to immune function were also expressed. Pit 1、 zinc finger protein 74 (ZNF74) and zinc finger protein PLAG1 were found with relative specific expression in pituitary. Conclusion Compared with the gene expression patterns of other tissues, e.g. heart or CD 34 + hematopoietic precursors, the percentages of genes involved in cell signal transduction/communication are significantly higher in normal pituitary, but similar to those ESTs data from brain, hypothalamus and adrenal gland. Galanin and CART are probably pituitary hormones related to the regulation of appetite. There are wide interactions between pituitary and immunological system. Pit 1、 ZNF74 and PLAG1 may be the transcription factors specifically expressed in normal human pituitary.
