摘  要:目的探讨垂体腺瘤中芳烃受体影响蛋白(AIP)基因的突变与垂体腺瘤侵袭性的相关性。方法选取病理证实的垂体腺瘤标本58例,按照Knosp分级分为侵袭性(26例)和非侵袭性(30例)垂体腺瘤,另选取正常成人尸头中垂体4例作为对照。对所有标本进行DNA提取、PCR扩增及DNA测序。结果 58例标本中AIP基因共发现3个突变位点,分别为Exon4中的一处C突变为T和Exon6中的两处G突变为C;28例侵袭性垂体腺瘤中有12例标本发生突变,突变率为42.8%;30例非侵袭性垂体腺瘤中有5例标本发生突变,突变率为16.7%;正常垂体组织中没有发现突变。侵袭性垂体腺瘤突变率明显高于非侵袭性垂体腺瘤(P<0.05)和正常垂体组织(P<0.05),而后两者无统计学差异(P>0.05)。结论侵袭性垂体腺瘤中AIP基因突变率明显高于非侵袭性垂体腺瘤,这提示AIP基因的突变与垂体腺瘤侵袭性存在相关性。Objective To investigate the relationship between the mutations of aryl hydrocarbon receptor interacting protein (AIP) gene and the invasiveness of pituitary adenomas in Chinese. Methods The pituitary adenomas tissues were collected from 58 patients with pituitary adenomas who underwent microsurgery from September, 2010 to September, 2012. Of 58 patients with pituitary adenomas, 28 were invasive pituitary adenomas with Knosp grade Ill ~ IV (invasive group) and 30 were non-invasive with Knosp grade 0~ II (non-invasive group). The normal pituitary tissues which were collected from 4 health adult cadaveric head were served as control group. The DNA was extracted from all the specimens and amplified by PCR, and then complete sequencing was performed on the DNA. Results The rate of AIP gene mutation in invasive group (42.9%, 12/28) was significantly higher than those in non-invasive group (16.7%, 5/30; P〈0.05) and control group (0; P〈0.05), but there were no significant difference between non-invasive group and control group (P〉0.05). One mutationai site with C to T mutation in Exon4 and two with G to C mutation in Exon6 were found in AIP gene of Chinese patients with pituitary adenomas, respectively. Conclusions The AIP gene mutation may be involved in the invasive growth of the pituitary adenomas. The mutation sites of AIP gene exist in Exon4 and Exon6 of AIP gene in Chinese patients with pituitary adenomas.
