摘  要:采用谷氨酰胺来确定氨基酸初始添加浓度,分析各种氨基酸对类弹性蛋白多肽[KV8F-40]产量的影响.结果表明:苏氨酸、精氨酸和半胱氨酸都能较大提高产量,其中以苏氨酸最为明显,产量可达126.9mg·L-1,比对照增加54.0%.利用响应面设计对上述3种氨基酸进行3因素3水平实验,经优化后所得理论最大产量为100.3mg·L-1.经实验验证,所得实验值为103.1mg·L-1,与理论值吻合,但其产量低于单个氨基酸.交互作用分析表明:3种对产量有促进作用的氨基酸之间没有互作或者存在拮抗作用.Using Gln to tentatively identify the initial concentration of each amino acid, we analyzed the influence of ami- no acids on the production of elastin-like polypeptides [KV8F-40]. The results showed Thr, Arg and Cys could improve the production at the concentration of 1.5 mmol L-1. Among them, Thr was the best; it could improve the production from 82. 4 nag L -1 to 126.9 mg L-1 , with an improvement of 54%. And then, we designed the response surface anal- ysis with three factors and three levels to optimize the cultivation medium of ELPs40. The experimental results fitted the theoretical result very well, which was 103. 1 mg L-1 and 100. 3 mg L-1 , respectively. However, the production was less than that of single factor. By means of interaction analysis, we found Thr and Cys inhibit each other, while others didn't have interactiorL
