摘 要:以丹皮酚的包合率为指标,比较了饱和溶液法和研磨法的优劣,并采用四因素三水平的正交试验研究不同体积的水研磨,β-环糊精:丹皮酚:挥发油比例,研磨时间及无水乙醇冲洗量对丹皮酚包合率的影响。采用HPLC以丹皮酚含量为标准,测定包合物中丹皮酚的包合率。得到了β-环糊精包合挥发油和丹皮酚的最佳工艺条件。最佳包合工艺为:β-环糊精:丹皮酚:挥发油=8:0.4:0.5,β-环糊精∶水=1:3,研磨2 h,用5 mL(加入0.5mL挥发油时)无水乙醇冲洗包合物。本方法包合率高,适合大量提取,可用于工业化生产。In this study, the paeonol inclusion rate was used as index to compare two inclusion methods, namely saturated water solution method and grinding method. In order to optimize the procedure, the effect of different volumes of lapping water,the ratio of β-cycledextrin/paeonol/volatile oil, lapping time and ethanol rinsing volume on the inclusion rate of paeonol were investigated by orthogonal experiment design (4 factors and 3 levels), the paeonol inclusion rate was deter- mined by HPLC using the content of paeonol as the standard. The optimal inclusion procedure was as follows : the ratio of β-eycledextrin/paeonol/volatile oil was 8: 0.4: 0.5, the ratio of β-cycledextrin/water was 3: 1,the grinding time was 2 h, rinsing volume of anhydrous ethanol was 5mL. The optimized procedure can provide high inclusion rate,and was easy for large amount extraction and industral production.
