摘  要:探讨大豆分离蛋白和低聚果糖联合应用对人免疫功能的调节作用。以不同浓度的大豆分离蛋白和低聚果糖混合物刺激正常人外周血单个核细胞(PBMC),分别测定淋巴细胞增殖、细胞因子分泌、B细胞分化和抗体产生等作用。不同浓度大豆分离蛋白和低聚果糖混合物刺激后均使淋巴细胞增殖能力增强(P<0.05),但明显低于ConA诱导的淋巴细胞转化作用;细胞因子IL-2、IL-12和IFN-γ的分泌水平明显高于未刺激组(P<0.05),且IL-12和IFN-γ的分泌水平呈现剂量依赖性;在中、低混合物浓度作用下,CD19+B细胞和CD19+CD138+B细胞的比例都较对照组有升高,而高剂量混合物刺激时该细胞比例变化不明显。研究结果还显示,和对照组相比,中浓度混合物刺激后IgG抗体的分泌水平明显增加(P<0.05),而低浓度混合物刺激时IgM抗体的分泌水平明显增加(P<0.05)。结果显示,大豆分离蛋白和低聚果糖联合作用可以在体外增强人体的细胞免疫和体液免疫功能,并且两种免疫功能在不同浓度的混合物联合作用下呈现不同的变化格局,提示两种成分对细胞免疫和体液免疫功能具有不同调节作用。To investigate the regulatory effects of soybean protein isolate/fructooligosaccharide mixture on human immune re- sponses in vitro, human peripheral blood mononuclear cells (PBMCs) were treated with soybean protein isolate/fructooligosac- charide mixture at different concentrations in vitro. Immune responses including ConA-triggered T lymphocyte proliferation a- bility, cytokine secretion, B lymphocyte differentiation and antibody production were evaluated. T lymphocyte proliferation a- bility was significantly elevated when stimulated by soybean protein isolate/fructooligosaccharide mixture at different concentra- tions (P〈0.05), but much lower than that upon ConA stimulation, The levels of cytokines IL-2, IL-12 and IFN-γ in the su- pernatant were remarkably higher after treatment by the mixture than those in non-treated groups (P〈0.05), and a dose-de- pendent increase in production of IL-12 and IFN-7 was obvious. Percentage of both CD19+ and CD19+ CD138+ B lymphocytes in total PBMCs increased when treated with low or medium concentration of the mixture, however, no obvious increase was ob- served in the group treated with high concentration of the mixture, Compared to the control group, IgG level was significantly upregulated in response to medium concentration of the mixture (P〈0.05) while the level of IgM in low-concentration-treated group was the highest (P〈0.05). Soybean protein isolate/fructooligosaccharide mixture thus exhibits enhanced immune regu- latory roles in vitro through modulating both cellular and humoral immune responses. As the response pattern varied with the individual concentraion of soybean protein isolate and fructooligosaccharide in the mixture, it would probably suggest that seg- regated immunomodulatory effects of soybean protein isolate and fructooligosaccharide on either cellular or humoral immunity.
