摘  要:目的研究NOB1反义寡核苷酸(NOB1-ASODN)对人卵巢癌细胞株SKOV3生长、凋亡的影响,观察NOB1-ASODN对NOB1基因表达的作用。方法设计合成针对NOB1 mRNA的特定的ASODN和作为对照的无义寡核苷酸链(NSODN),应用转染载体X-tremeGENE siRNA Transfection Reagent将ASODN及NSODN转染至人卵巢癌SKOV3细胞。采用RT-PCR法检测转染后各组细胞NOB1 mRNA的表达;应用MTT法检测不同作用时间段SKOV3细胞的生长情况;流式细胞学检测细胞的凋亡情况。结果 NOB1-ASODN能下调SKOV3细胞NOB1基因的表达,沉默效率达59.58%;其能抑制细胞的生长,具有时间依赖性,转染后24、48、72 h细胞抑制率分别为(37.02±0.710)%、(56.16±0.986)%、(58.10±0.010)%;沉默目的基因后细胞的凋亡率增加。结论运用NOB1-ASODN特异、有效地抑制NOB1表达,能抑制SKOV3细胞的增殖,可促进SKOV3细胞的凋亡,从而达到抑制肿瘤生长的目的。Objective To study the effects of NOB 1 antisense oligonucleotide ( ASODNs ) on cell growth and apoptosis of human ovarian cancer cell, and observe the effects on expression of NOB1 gene. Methods The specific ASODNs for NOB1 mRNA and the nonsense oligonucleotide chain NSODNs as a control were designed and synthesized. Then ASODNs and NSODNs were infected into SKOV3 cells by the transfection carrier X-tremeGENE siRNA Transfection Reagent. RT-PCR was used to detect the expression of NOB1 mRNA after the transfection in each group cells ; MTF assay was used detect the SKOV3 ' s growth in different time segments ; Flow cytometry was performed to detect the apoptosis on SKOV3. Results NOB1-ASODNs could down regulate the expression of NOB1 gene at mRNA (59.58%) ; inhibit the growth of cells in a time-dependent manner after transfection, cells inhibition rate on 24 h : (37.02 ± 0. 710) %, 48 h : (56. 16 ± 0. 986) %, 72 h : (58.10± 0.010) % ; increase the rate of apoptosis. Conclusion NOB1-ASODN can specifically and effectively inhibit the expression of NOB1 gene, inhibit the proliferation of SKOV3 cells and promote apoptosis of SKOV3 cells, so as to achieve the purpose of inhibiting tumor growth.
