摘 要:目的探讨肾癌组织中胰岛素生长因子Ⅱ(IGF-Ⅱ)的印记丢失状态及意义。方法应用限制性内切酶切片段长度多态性分析法,检测印记基因IGF-Ⅱ在75例肾癌患者的癌组织及其癌旁组织中的印记状态。结果在75例肾癌标本中,癌及癌旁组织IGF-Ⅱ均为杂合子基因型的58例(77.3%)。其中有26(44.8%)例癌及癌旁组织均发生IGF-Ⅱ印记丢失(LOI);5(8.62%)例癌组织发生IGF-ⅡLOI而癌旁组织IGF-Ⅱ印记正常;1(1.72%)例癌旁组织发生IGF-ⅡLOI而癌组织为正常印记状态。在14岁到72岁的各年龄段男性和女性来源组织标本中均存在癌及癌旁组织IGF-ⅡLOI的发生,且频率相近,没有统计学差异。在58例杂合子组织标本中,分别有31(53.4%)例癌组织、27(46.6%)例癌旁组织发生IGF-ⅡLOI。统计分析结果表明肾癌癌组织IGF-ⅡLOI发生率与癌旁组织IGF-ⅡLOI发生率没有统计学差异。结论肾癌癌组织及癌旁组织IGF-ⅡLOI的发生与性别及年龄无显著相关,肾癌中癌旁组织IGF-ⅡLOI的发生率与癌组织接近,可能是疾病发生早期事件。Objective Exploring the status and significance of loss of imprinting (LOI) of IGF-1I gene in renal cell carcinoma. Methods Restriction fragment length polymorphism (RFLP) analysis were used to detect the imprinting status of IGF- II gene in 75 renal cell carcinoma tissues and matched histological normal kidney tissues. Results There were 58 renal cell carcinoma cases with heterozygous IGF-11 gene (77.3%, 58/75). In these 58 cases, 26 cases had loss of imprinting (LOI) of IGF-lI gene both in carcinoma tissues and matched histological normal kidney tissues (44.8%,26/58), and 5 cases had IGF II LOI in carcinoma tissues (8. 62%,5/58), and 1 cases had IGF-II LOI in matched histological normal kidney tissues (1.72%, 1/58). IGF-II LOI occurred on both genders and all ages, there was no statistical differences. In 58 heterozygous cases, there were 31 (53.4% ,31/58) and 27(46.6%) cases had IGF- II LOI in renal carcinoma tissues and matched histological normal kidney tissues, respectively. Statistical analysis showed that there was no statistical significant between the IGF- II LOI in renal cell carcinoma tissue and the matched histological normal kidney tissues. Conclusion There is no significant correlation between IGF- II LOI and the gender and age of renal cell carcinoma patients. IGF II LOI incidence is close in renal cell carcinoma tissues and matched histo logical normal kidney tissues, which indicates that IGF-II LOI may occurs in the early stage of the disease.
