流式细胞术分析牛乳铁蛋白素促进Jurkat细胞早期凋亡

摘　　要：牛乳铁蛋白素(Bovine Lactoferrcin,LfcinB)是一类具有广谱抗菌作用的生物活性肽,能够选择性诱导肿瘤细胞凋亡,但并没有对LfcinB诱导肿瘤细胞的凋亡特性进行分析。本实验利用荧光倒置显微镜初步检测牛乳铁蛋白素(LfcinB)影响Jurkat细胞增殖的作用方式,运用Annexin V-FITC/PI双标记流式细胞术将LfcinB促进Jurkat细胞凋亡的阶段进行区分和定量。通过实验得出结论,LfcinB是通过诱导Jurkat细胞发生早期凋亡来抑制其增殖的,并且早期凋亡率与作用浓度呈正相关。这一结论为更深一步研究LfcinB对肿瘤细胞的作用机制奠定基础。Bovine lactoferrcin （LfcinB） could induce apoptosis of cancer cells as biological active peptide for the effect of broad-spectrum antibacterial. However,it was rarely to analyse the apoptosis characteristics of LfcinB induce cancer cells. In this experiment,inverted flurescence microscopy was used to initial survey the mode of action from LfcinB effect to Jurkat cells,also distinguished and quantified the apoptosis stage of LfcinB effect to Jurkat cells. According to these results,the suppress proliferation of Jurkat cells was caused by LfcinB induce cells early apoptosis,and the early apoptosis ratio was positively related to the action concentration. These results were bases to research the mechanism of LfcinB induce cancer cells.
