摘  要:目的研究γ射线照射对含KDR启动子双自杀基因大肠癌细胞SW480的杀伤协同作用,和自杀基因及前药的放射增敏作用。方法重组腺病毒pAdKDR-TK、pAdKDR-CD和pAdKDR-TK/CD转染SW480细胞,观察γ射线对SW480细胞基因转染率的增强作用和对前药GCV、5-FC、GCV+5-FC杀伤SW480细胞的放射增敏效应,以及前药GCV、5-FC、GCV+5-FC对γ射线杀伤SW480细胞的协同增效作用。结果转染双自杀基因的大肠癌细胞射线组比对照组转染率有明显升高(P<0.01),γ射线对转染KDR-TK/CD基因的SW480细胞比未转染基因的对照组SW480细胞具有更强的杀伤作用(P<0.001)。结论γ射线照射能明显提高双自杀基因的转染效率,强化了基因治疗的"旁观者效应",自杀基因对γ射线的增敏作用。Objective To investigate the killing effect to colorectal SW480 cells of the co-expression adenovirus vec- tor containing double suicide genes. Utilize the double suicide genes radiated by y-ray to observe it's co-inhibition role and double suicide genes and prodrug's radiosensitization. Methods Recombinant plasmid pAdKDR-TK,pAdKDR-CD and pAdKDR-TK/CD infected SW480 cells, The increasing function of y-ray on gene-modified rate of colorectal SW480 cells,the radiosensitizing effect of y-ray to prodrug GCV,5-FC and GCV+5-FC on colorectal SW480 cells,and the co- operated synergistic action of prodrug GCV,5-FC and GCV+5-FC on colorectal SW480 cells with y-ray radiation were observed. Results The infected with TK/CD double suicide gene radiated by )'-ray had higher infected rate than that of control group which had noy-ray radiation (P〈0.01);SW480 cells infected with double suicide gene radiated by y-ray was more sensitive than the control group (P〈0. 001). Conclusion The )'-ray can increase the rate of transfection of SW480 cells obviously,enhance the killing effect to cells for prodrug,and had radio-sensitizing effect.
