摘 要:目的分析乳腺癌17号染色体多体在HER-2基因状态及临床病理特征中的意义。方法应用荧光原位杂交(FISH)技术检测186例浸润性乳腺癌患者石蜡切片中HER-2基因扩增与17号染色体多体情况,并分析17号染色体多体与HER-2基因扩增及各临床病理特征的关系。结果 186例样本中,17号染色体多体比例为43.0%(80/186),其中HER-2基因扩增组66.3%(53/80),HER-2基因未扩增组25.5%(27/106),前者显著高于后者(x2=30.929,P=0.000);17号染色体多体与肿瘤大小及肿瘤分级显示相关(r=-0.188,P=0.001;r=0.159,P=0.03),而与年龄、Ki67、淋巴结转移﹑ER、PR表达及ER/PR状态无关。结论 17号染色体多体与HER-2基因扩增者相关;并且可能提示乳腺癌患者预后不良。Objective To analyze the significance of 17th chromosome polysomy of breast cancer in the HER-2 gene statu and clinical pathological features. Methods The HER-2 gene amplification and 17th chromosome polysomy in paraffin sections of 186 patients with invasive breast cancer were detected by using fluorescent in situ hybridization(FISH), and the relationships between 17th chromosome polysomy and HER-2 gene amplification and between 17th chromosome polysomy and clinical pathological features were analyzed. Results The proportion of 17th chromosome polysomy was 43.0% (80/186)among 186 individuals, of which the proportion of 17th chromosome polysomy in HER-2 gene amplification group was 66.3% (53/80), the proportion in HER-2 gene non-amplification group was 25.5%(27/106), the proportion of the former was significantly higher than that of the latter(x2=30.929, P=0.000); The 17th chromosome polysomy was associated with tumor size and tumor grade(r=-0.188, P=0.001; r=0.159, P=0.03), but was not associated with age, Ki67, lymph node metastasis, expressions of ER and PR, ER/PR. Conclusion The 17th chromosome polysomy is associated with HER-2 gene amplification; And it can indicate poor prognosis in patients with breast cancer.
