摘  要:目的探讨不缺硒、高脂膳食情况下,进一步补充硒对血脂水平、血浆氧化应激和脂质过氧化的影响。方法40只雄性大鼠随机分为对照组、单纯高脂饲料(HFD)组,高脂饲料加硒(HFD+Se+)组和高脂饲料加高硒(HFD+Se++)组。对照组、HFD、HFD+Se+和HFD+Se++组饲料中硒含量为0.3 mg/kg、0.3 mg/kg、0.6 mg/kg和1.2 mg/kg。16周后处死大鼠,测定血浆硒浓度、血脂水平(TG、TC、HDL-C、LDL-C)、氧化应激和脂质过氧化水平(GPx、SOD、MDA)。结果高脂喂养的大鼠体重和腹腔脂肪系数高于对照组(P﹤0.01),高脂喂养导致大鼠脂代谢异常和抗氧化能力受损。补硒进一步升高血硒水平,HFD+Se++组大鼠血硒水平高于对照组(P﹤0.01)。饲料硒含量不同的3组高脂喂养大鼠之间的血脂水平、血浆GPx活性无显著差异,但HFD+Se+和HFD+Se++组大鼠的血浆MDA含量低于HFD组(P﹤0.05)。结论硒不缺乏情况下进一步补硒不能改善高脂喂养所致的脂代谢异常,但部分降低了脂质过氧化。OBJECTIVE The study focused on investigating the effect of selenium supplements on blood lipid level, plasma oxidative stress, and lipid peroxidation in rats fed a selenium-sufficient and high-fat diet. METHODS 40 male Wistar rats were randomly divided into 4 groups, including a control group, a high-fat diet (HFD) group, a selenium-supplemented high-fat (HFD Se+) group, and a high-fat high-selenium (HFD+Se++) group; selenium contents for the 4 groups were 0.3 mg/kg, 0.3 mg/kg, 0.6 mg/kg, and 1.2 mg/kg, respectively. The rats were sacrificed at the end of 16 weeks, and plasma levels of selenium, lipids (TG, TC, HDL-C, and LDL-C), oxidative stress, and lipid peroxidation (GPx, SOD, and MDA) in the rats were determined. RESULTS High fat diets were shown to induce higher body weight and higher peritoneal adiposity as compared with the control group (P 〈 0.01 ), resulting in dyslipidemia and impaired antioxidative function in the rats. Selenium supplements increased plasma selenium level; thus, HFD+Se++ rats had higher plasma selenium level than the control group (P 〈 0.01 ). No significant difference in blood lipid level and plasma GPx activity was observed between the 3 groups of high-fat diets with different selenium contents; however, plasma MDA contents were significantly lower in the HFD+Se++ and HFD+Se++ groups than in the control group (P 〈 0.05). CONCLUSION Selenium supplementation under sufficient selenium intake did not show to improve high-fat diet-induced dyslipidemia, but was able to partially reduce lipid peroxidation.
