摘 要:研究复合酶解法提取虎杖中白藜芦醇的最佳工艺条件。在单因素试验基础上,以水作为溶剂,固液比1∶20(g/mL),选取酶添加量、酶解pH、温度和时间4个因素,采用L9(34)对酶解法提取白藜芦醇工艺条件进行正交试验,得到最佳提取条件为,复合酶添加量为1.5 mg/g纤维素酶+1.5 mg/g阿魏酸酯酶,温度50℃,pH 5.0,提取时间3.0 h,白藜芦醇提取率为1.513%。与溶剂提取法、纤维素酶酶解法相比较,复合酶解法提取虎杖中白藜芦醇的效率明显提高。To study the extraction optimum process conditions of resveratrol in Polygonum cuspidatum by composite enzymatic hydrolysis. Enzyme hydrolysis method was used the various factors influencing extraction through orthogonal tests designed L9(34) to find the best conditions on the single factor experiment studied. The result showed that the optimized condition be that composite enzyme dosage was 3 mg/g (that was 1.5 mg/g cellulose and 1.5 mg/g ferulic acid esterase), temperature 50 ℃, pH5.0, time of enzymatic hydrolysis 3 h on solid-liquid ratio: 1:20 (g/mL, water as solvent)). The acquired amount of resveratrol in Polygonum c uspidatum extraction was about 1.513 %. The resveratrol extraction result by enzymatic hydrolysis method was obviously better than the other extraction method in optimized condition.
