金水鲜胶囊联合放射治疗对肺腺癌A549细胞HIF-1α和EGFRmRNA表达的影响

摘　　要：[目的]研究金水鲜胶囊联合放疗对肺腺癌A549细胞增殖的抑制作用以及HIF-1α和EGFR mRNA表达的影响。[方法]将处于对数期生长的A549细胞随机分为4组(空白对照组、放射治疗组、金水鲜胶囊组和放疗治疗+金水鲜胶囊组),MTT法计算细胞生长抑制率,RT-PCR技术检测HIF-1α和EGFR mRNA的含量变化。[结果]各组肺腺癌A549细胞生长受到不同程度的抑制,以金水鲜胶囊联合放疗组最明显,差异有统计学意义(P<0.05)。除放疗组外,各组HIF-1α和EGFR mRNA表达均下降。48h时,金水鲜胶囊组及金水鲜胶囊联合放疗组与空白对照组、放疗组相比,HIF-1α和EGFR mRNA表达差异有统计学意义(P<0.05)。[结论]金水鲜联合放疗可抑制肺腺癌细胞增殖,下调HIF-1α和EGFR mRNA的表达。[Purpose] To investigate the inhibitory effect and the expression of HIF-1α mRNA and EGFR mRNA of Jinshuixian capsule combined with radiotherapy in lung adenocarcinoma cell line A549.[Methods] A549 cells in log phase were randomly divided into 4 groups：control group,radiotherapy group,Jinshuixian group and Jinshuixian plus radiotherapy group.The cell growth inhibition rate was evaluated by MTT method,and HIF-1 α and EGFR mRNA were detected by RT-PCR technique.[Results] The cell growth in lung adenocarcinoma cell line A549 was inhibited with the significant inhibition in Jinshuixian plus radiotherapy group（P 0.05）.In addition to radiotherapy group,expression of HIF-1 α and EGFR in other group was decreased.The level of HIF-1α and EGFR mRNA was significant different in Jinshuixian group,Jinshuixian combined radiotherapy group compared to control group and radiotherapy group（P all 0.05）.[Conclusion] Jinshuixian combined with radiotherapy could inhibit the proliferation of lung adenocarcinoma cells,and decrease the expression of HIF-1 α and EGFR mRNA.
