摘  要:为确证蛹虫草虫草素的抗肿瘤功效,研究虫草素对人乳腺癌细胞MCF-7和小鼠S180肉瘤的抑制作用效果。采用四甲基偶唑蓝(MTT)法和流式细胞术检测人乳腺癌细胞MCF-7的增殖与凋亡情况,结果表明:0.1~5.0 g/L虫草素可显著抑制人乳腺癌细胞MCF-7的生长,并呈时间剂量依赖性;0.8 g/L虫草素处理24、48、72 h后,MCF-7细胞的生长抑制率与凋亡率分别为32.6%、53.3%、87.6%和8.65%、14.20%、26.30%,处理72 h后G0~G1期细胞比率显著增加。S180肉瘤小鼠灌胃给予虫草素10 d后,高、中、低剂量组[90、60、30 mg/(kg·d)]小鼠体内的S180肉瘤生长受到明显的抑制(P<0.01),抑制率分别为64.33%、44.59%、32.48%,而且具有一定的量效关系;中、低剂量虫草素能增加S180肉瘤小鼠的脾脏指数和胸腺指数。试验结果证明,虫草素能够有效抑制人乳腺癌细胞MCF-7增殖和小鼠S180肉瘤的生长以及提高小鼠的免疫能力。In order to confirm the antitumor effect of Cordyceps mifitaris cordycepin, inhibitive effects of cordycepin on human breast cancer cell MCF-7 and Slso-bearing mice were investigated. The proliferation and apoptosis of MCF-7 cells were measured using methyl thiazolyl tetrazolium (MTT) and flow cytometry. The results showed that cordycepin with mass concentrations ranging from 0. 1 to 5, 0 g/L could inhibit proliferation of MCF-7 cells significantly and the inhibition was in a time- and dose-dependent manner. After treatment with 0.8 g/L cordycepin for 24, 48 and 72 h, the growth in- hibition rate of MCF-7 cells was 32.6%, 53.3% and 87.6% respectively, and the apoptosis rate of MCF-7 cells was 8.65%, 14.20% and 26.30% respectively. Moreover, the proportion of cells at GO to G1 phases was increased signifi- cantly after treatment for 72 h. The growth of S180 sarcomas in mice was extremely significantly inhibited ( P 〈0.01 ) after treatment with high, medium and low dosages [90, 60 and 30 mg/(kg·d) ] of cordycepin. The inhibition rate was 64.33%, 44.59% and 32.48% respectively, which was dose-dependent as well. Medium and low dosages of cordyce- pin could increase spleen index and thymus index of S180-bearing mice. These results confirmed that cordycepin can ef- fectively inhibit the proliferation of MCF-7 ceils and the growth of S180 sarcoma and increase the immunity of mice.
