摘 要:以樟树籽油(Camphor seed oil,CSO)为原料,优化脂肪酶水解CSO制备二酰甘油(diacylglycerol,DAG)的工艺。通过丙酮和叔丁醇对脂肪酶进行预处理,对3种固定化脂肪酶(Lipozyme RM IM、Lipozyme TL IM、Novozym 435)进行筛选。以反应产物DAG质量分数和三酰甘油(TAG)质量分数为响应值,含水量(加酶量的30%~50%)、加酶量(油质量的8%~10%)和温度(55~75℃)作为变量,利用响应面法(RSM)优化CSO制备DAG工艺。结果表明,Lipozyme RM IM经过预处理后,重复使用次数可达到11次,RSM法优化CSO制备DAG的最优工艺条件:加酶量10%,含水量40%,温度65℃。在此优化条件下,DAG质量分数和TAG质量分数分别为50.38%和40.68%。此工艺为樟树籽油工业化生产中链DAG油提供理论和试验依据。With camphor seed oil (CSO) as raw material, the craft of CSO to produce diacylglycerol (DAG) under the lipase hydrolysis was optimized. With pretreatment of organic solvents acetone and tert - butanol, the best lipase was chose among three commercial immobilized lipases including Lipozyme RM IM, Lipozyme TL IM and Novozyme 435. Using quantity scores of DAG and Triacylglycerol (TAG) as the response values, and setting the water content ( 30 % -50 % of enzyme mass), enzyme content ( 8 %- 12 % of oil mass) and reaction temperature ( 55 - 75 ℃) as the variables,the response surface methodology( RSM)was applied to optimize the process of producing DAG by hydroly- sis of CSO. The results showed that Lipozyme RM IM could be reused for 11 times after being pretreated. The RSM optimal reaction conditions of DAG were :40% of water content, 10% of enzyme load ,65 ℃of reaction temperature, and under this condition, the quantity scores of both DAG and TAG were 50.38% and 40.68 %, respectively. This craft can provide theory and test values for the application of Commercialized production of the Medium Chain DAG from CSO.
