摘 要:目的通过比较ARMS实时荧光定量PCR法和DNA直接测序法,分析K-ras基因突变检测结果的一致性,确立临床K-ras基因突变检测的最适方法。方法应用ARMS实时荧光定量PCR法及DNA直接测序法检测34例石蜡标本的K-ras基因第12位和第13位密码子的基因突变,结果比较采用χ2检验。结果这两种方法对K-ras基因第12和第13密码的基因突变检测结果无统计学差异,ARMS实时荧光定量PCR法比测序法操作简便,敏感度更高。结论相较于DNA直接测序法,ARMS实时荧光定量PCR法更适用于临床检测。Objective To compare the two methods of ARMS real-time PCR and DNA sequencing in detection of K-ras mutations. Methods K-ras 12th and 13th codon gene mutation were detected by ARMS real-time PCR and DNA sequencing in 34 paraffin specimens. Chi-square test was used to analyze the consistance. Results There was no significant difference between the two methods. However, ARMS real-time PCR was more convenient and sensitive compared to DNA sequencing. Conclusion ARMS real-time PCR is more suitable for clinical testing than DNA sequencing.
