摘 要:目的:研究胃癌组织中RASAL1基因启动子的甲基化状态并分析其与临床病理资料的关系,初步探讨RASAL1基因启动子甲基化在胃癌发生发展中的意义。方法:取40例胃癌患者手术标本,使用甲基化特异性PCR(MSP)技术分别检测胃癌组织及相应癌旁组织中RASAL1基因启动子的甲基化状态,并分析其与胃癌临床病理特征的关系。结果:胃癌组织和癌旁组织中RASAL1基因启动子甲基化率分别为70%(28/40)和30%(12/40),胃癌组织中RASAL1基因启动子的甲基化发生率明显高于癌旁组织,P<0.01。胃癌组织中RASAL1基因启动子的甲基化率与性别(P=0.622)、年龄(P=0.168)无明显相关;但与分化程度(P=0.006)、肿瘤大小(P=0.043)、侵袭深度(P=0.015)和淋巴结转移相关(P=0.010);在肿瘤体积大、分化程度差、侵袭深度较深和有淋巴结转移的胃癌组织中,RASAL1基因启动子甲基化率明显增高。结论:胃癌组织中RASAL1基因启动子区域甲基化发生率高,且与分化程度及进展阶段相关,提示RASAL1基因启动子的异常甲基化可能参与了胃癌的发生发展过程。OBJECTIVE:To detect the methylation status of RASAL1 gene promoter in gastric cancer tissues, and analyze the relationship between clinicopathologie features and the RASAL1 promoter methylation,so as to study the role of promoter hypermethylation of RASAL1 gene in the course of tumorigenesis and progression in gastric cancer. METH- OD: Methylation-specific polymerase chain reaction (MSP) was used to detect RASAL1 promoter methylation in tumor and tumor-adjacent tissues from 40 gastric cancer patients and the relationship beteewn the clinicopathologic features and the methylation frequency of RASAL1 promoter was analyzed. RESULTS: The frequency of RASAL1 promoter methyla- tion in gastric cancer tissues and tumor-adjacent tissues were 70 % (28/40) and 30 %(12/40) respectively (P〈0.01). No correlation was found between RASAL1 promoter methylation and age(P=0. 622) or gender(P=0. 168). There was sig nificant correlation between the frequency of RASAL1 promoter methylation and tumor differentiation(P: 0. 006), tumor size(P=0. 043) ,invasive depth(P=0. 015) and lymph node metastasis(P=0. 010) in patients with gastric cancer. CON- CLUSION: The aberrant promoter methylation of RASAL1 gene is a common event in gastric cancer, and it may play a role in the tumorigenesis and progression of gastric cancer.
