摘  要:目的探讨番茄红素对鱼藤酮诱导的PC12细胞凋亡的作用及可能机制。方法采用鱼藤酮诱导PC12细胞为损伤模型,在番茄红素预处理后,以CCK-8法检测细胞存活率,Hoechst33258染色荧光显微镜观察细胞凋亡形态学改变,流式细胞术检测细胞凋亡率,罗丹明123染色检测细胞线粒体膜电位,比较各组的差异。结果与鱼藤酮组相比,番茄红素预处理后,细胞存活率明显提高(P<0.05),细胞形态明显改善,凋亡率降低(P<0.05),线粒体膜电位上升(P<0.05)。结论番茄红素对鱼藤酮诱导的PC12细胞凋亡具有抑制作用,其机制可能与其稳定线粒体膜电位有关。Objective To investigate lycopene on rotenone induced apoptosis in PC12 cells and its possible mecha- nism. Methods Rotenone-induced PC12 cells were used in the study. Cell viability was determined by CCK-8 assay,The morphological change of PC12 cells was observed by Hoechst33258 staining, The apoptosis was monitored by FCM, Mito- chondrial membrane potential(MMP) of PC12 cells was detected by Rh123 staining. Results Lycopene significantly atten- uated the cytotoxic effect of rotenone. It increased the cell survival as measured with CCK-8, decreased the apoptosis rate, and inhibited MMP collapse. Conclusion Lycopene has a possible neuroprotective role in rotenone-induced apoptosis of PC12 cells, and the mechanism of the effects of lycopene may be involved in inhibiting MMP collapse induced by rotenone.
